There's a good chance the biography is wrong. Both Bonanno and Costello insisted the were personally engaged in bootlegging with Kennedy.

I think the weight of the evidence is that he was involved. As to the degree, or to what extent that activity accounts for the family fortune, I guess reasonable persons could differ.
